Isoelectric analysis of cytolytic bacterial proteins.

چکیده
In this note, we describe the application of isoelectric focusing to the characterization of two cytolytic proteins, the a-toxin or phospholipase C of Clostridium perfringens (clostridial PLC) and cereolysin. We used a 110-ml electrolysis column (LKB Instruments, Rockville, Md.), the design of which was given by 0. Vesterberg et al. (Biochem. Biophys. Acta 133:435, 1967), who also summarized the theoretical and practical filtrate of C. perfringens Type A, kindly supplied by R. 0. Thomson (Weilcome Research Laboratories, Beckenham, Kent, England), and with the same conditions (without a pH gradient) used by Bangham and Dawson, but substituting sucrose for glycerol, we obtained (Fig. 1) a single peak of phospholipase activity, as measured by the capacity of fractions to cause turbidity in diluted egg yolk.

Immunogens of Brucella Abortus S19 Identified By Two-Dimensional Gel Electrophoresis and Immunoblotting
Background: Lipopolysaccharides (LPSs) and several antigenic proteins of Brucella have been considered for preparation of diagnostic reagents and subunit vaccines. The objective of this study was to identify and compare immunogens of B. abortus S19 which induce humoral immune responses in human, goat and rabbit. Material and Methods: The bacterial whole cell extract was prepared in extraction b...
متن کاملChemical modification of equinatoxin II, a lethal and cytolytic toxin from the sea anemone Actinia equina L.
The role of arginine and tyrosine in cytolytic properties of equinatoxin II, isolated from the sea anemone Actinia equina L., was studied by means of chemical modifications. The toxin was modified with 2,3 butanedione and tetranitromethane, respectively. The extent of modification and physico-chemical properties of the modified proteins were checked with amino acid analysis, isoelectric focusin...
Background: The non-enzymatic glycosylation (NEG) of proteins in diabetes damages both the structure and function of these proteins. In vivo and in vitro studies have shown that NEG of proteins and advanced glycosylation end-products (AGE) contribute to the pathogenesis of both macrovascular, such as atherosclerosis, and microvascular complications, such as retinopathy and nephropathy, in diabe...

An extracellular cytolytic toxin produced by the halophilic bacterium Vibrio vulnificus was isolated free of detectable contamination with medium constituents and other bacterial products by sequential ammonium sulfate precipitation, gel filtration with Sephadex G-75, hydrophobic interaction chromatography with phenyl-Sepharose CL-4B, and isoelectric focusing in an ethylene glycol density gradi...
متن کاملOptimum conditions for protein extraction from tuna processing by-products using isoelectric solubilization and precipitation processes
The by-product from tuna processing is a potential source of edible protein. Therefore, it is very important to extract protein from such raw materials for human food. In this study the optimum pH for protein extraction from tuna by-products was optimized by using isoelectric solubilization and precipitation processes. The Response Surface Methodology (RSM) and the single factor model were used...
